I am fairly new to Knoppix (as well as being begginer to advance use/hacking of Linux), which is wonderfull Debian based Live CD distro, so I can only promisse to report when I get it running... if someone does it first please report!

This should be almost the same as using
TWikiOnDebian, since there's a .deb package that should simplify things significantly. Of course, you'll need to install the data and config files on the hard disk, which may complicate things - may be best to just install Knoppix to the HD first before doing TWiki installation.
You could even put together a bunch of PCs using
Google:ClusterKnoppix and see if it speeds things up

I do not think it will be easy to persuade Knopix developers to squeeze Twiki onto the CD. Twiki does not run 100% on plain Debian yet.

Space on single CD is very limited, competition fierce. They even use advanced (beyond unstable) code to make Knoppix happen, see many horror stories about twaht happend after next upgrade.
If you want easy-to install wiki, try
KwikiWiki. Anybody can install Kwiki in 15 minutes max, guaranteed. Even I did it

Of course not all the features of Twiki - yet.

I think it's probably enough to make sure that TWiki is easily installed on Knoppix-based systems, rather than trying to get it on the CD - after all, anyone using Knoppix on a server will need to install it to hard disk first. Since packages tend to grow over time, Klaus Knopper tends to have to drop packages in every new release to make room for bloat, rather than adding new packages...
I'm going to install Knoppix on my home PC and use it for TWiki development, so I'll see how I get on with
TWikiOnDebian. See
