Please let me know, how can i take backup of my twiki site.
What will be the restore procedure, if the machine crashes.

All the TWiki data is always stored fully on disk. So just backing up the TWiki installation dir + the apache config file + the crontab lines to start housekkeping (email notify) processes) is sufficient. Nothing special to restore, just re-put the dir in place + apache config + crontab and everything will work, no need to start.
Note that backuping the Twiki install dir is quite enough. the rest can be redone by hand like a first install.
