The TWikiOnMemoryStick using MicroWeb appears to me to be a program you can run without having to boot an OS, while PuppyOS appears to require booting your whole PC from the memory stick with the PuppyOS distro installed (if it's on a memory stick). That's just what I could tell from an initial look at the PuppyOS documentation though... is this pretty much the case or am I missing something?

What I've got working at the moment is the following :
sit down at a cyber cafe
insert memory stick into any computer
bootup your PuppyOS from the stick by running a .bat
shutting down the OS prompts to save changes to an .iso, but I don't have that part going yet
at no time are you prompted to install any software or reboot the host PC
My purposes behind putting the PuppyOS on a memory stick were not at all TWiki related. But the puppy is serving my other needs from a very small footprint. Once I get the read-write functionality to the USB along with a robust SyncContrib, then I'd probably also look into having a mini-twiki in there, as well.
